They have put a cordon around the area, evacuated nearby buildings and asked people to stay away.
Detective Chief Superintendent Russ Jackson said of the vehicle: 'This is potentially a significant development in the investigation'.
The auto is a white Nissan Micra and police are appealing for anyone who knows about its movements in the last few months to come forward.
The Nissan Micra was located close to Manchester Royal Infirmary, although the hospital remains open as normal.
The developments came Friday after police have released new security camera images of the Manchester bomber's last moments, hoping to piece together his final preparations before the concert blast.

The man was arrested today on suspicion of offenses contrary to the Terrorism Act after police cordoned off an area in southern Manchester visited by the bomber, Salman Abedi.
