Leopoldo Lopez and Antonio Ledezma were both under house arrest, the former for his role in leading street protests against President Nicolas Maduro in 2014 and the latter on charges of plotting a coup.
She included a link to a video of what appears to be Lopez being led into a vehicle emblazoned with the word Sebin, Venezuela's intelligence agency.
His wife, Lilian Tintori, wrote on Twitter that Mr López had been taken from his home at 12:27 local time (04:27GMT). "We don't know where he is or where they're taking him".
Mr López was transferred from a military prison to house arrest on 8 July after spending more than three years in jail.
A woman can be heard shouting "They're taking Ledezma, they're taking Ledezma, dictatorship!"
Mr Ledezma, a former Caracas mayor, was also detained in 2015 and has been under house arrest.
Maduro said Monday evening he had no intention of deviating from his plans to rewrite the constitution and go after a string of enemies, from independent Venezuelan news channels to gunmen he claimed were sent by neighboring Colombia to disrupt the vote as part of an worldwide conspiracy led by the man he calls "Emperor Donald Trump".
They don't intimidate me.
"I don't take imperialist orders, I don't take orders from foreign governments", he added. "Bring on more sanctions, Donald Trump". We won't accept an illegitimate government.
Mr Maduro has said the new assembly will begin to govern within a week.
Maduro was added to a list of individuals subject to sanctions for undermining democracy in Venezuela, freezing any assets under USA jurisdiction and barring US citizens or businesses from doing business with him.
Before their arrest, the men had called for mass protests against President Maduro's decision to create a new Constituent Assembly to change the country's Constitution.
